ABSTRACT: We characterized the benthic structure of the Guano Islands, Islets, and Capes National Reserve – Punta Coles in February and October 2019. A stratified design was applied (Stratum I: 0 – 5 m, Stratum II: 5 – 10 m, and Stratum III: 10 – 15 m), establishing six transects perpendicular to the coast and their replicas (n=3) with 18 stations. In order to collect macrobenthos, we used quadrants of 0.0625 m2 (destructive sampling) and gridded quadrants of 0.25 m2 and 1.0 m2 for sessile organisms and those greater than 2 cm in length (non-destructive sampling). Information was processed with the PRIMER V6.0 software. A total of 18 species were identified in the megabenthos, the phyla Echinodermata and Mollusca were more representatives, as well as the species Tetrapygus niger, Loxechinus albus, and the algae Lessonia trabeculata. Regarding coverage of organisms, 34 species were identified, and the phylum Mollusca had the greatest contribution to the species composition. The most representative taxa were Lithothamniom sp. and Balanus laevis. Pyura chilensis was representative in strata I and II and Aulacomya atra had a distribution restricted to the deepest stratum (>10 m). In macrobenthos, a total of 114 species were identified, being the phyla Mollusca, Arthropoda, and Annelida important in terms of abundance and species composition. The polychaete Syllis sp. and A. atra were the most important taxa in strata I – II and III, respectively. Shannon values (H’) were less than 1.5 bits/ind., and richness values per stratum varied between 64 and 75 species. nMDS analyzes indicated community clustering, based on two depth strata (0 to 10 m and >10 m). Environmental conditions indicated normal conditions in February and slightly warm in October. During the study period, Cold Coastal Waters (CCW) predominated.
